ABSTRACT

Objective To explore changes of gray matter volume in patients with obsessivecompulsive disorder (OCD) in Chinese Han population using optimized voxel-based morphometry (VBM) ,and investigate its relationship with clinical symptoms. Methods Twenty patients with OCD and 20 age,sex and handedness matched healthy controls were scanned using 3D-T1 images on a 3.0 T MR system. The high resolution T1WI was preprocessed according to the optimized VBM protocol in Statistical Parametric Mapping (SPM5). Two-sample t test was performed to characterize the differences of the gray matter volume (GMV) between the OCD patients and healthy controls, and the correlations between the GMV and symptom severity and cumulative illness duration were examined using Pearson correlation in SPSS 16. 0, respectively. Results Compared to controls, OCD patients demonstrated increased GMV in left thalamus, right thalamus and left cerebellum after false discovery rate (FDR) correction. No areas of significantly decreased GMV was observed in OCD patients in relative to healthy controls. The mean eigenvalue ranged from 0. 5782 to 0. 889 representing the left thalamus volume of OCD patients was 0. 6813 ± 0. 0718, and that ranged from 0. 5546 to 0. 9062 was 0. 6869 ± 0. 0808 tor right thalamus. The mean eigenvalues were positively correlated in bilateral thalamus (r = 0. 94, P < 0. 01). Conclusion Using optimized VBM, the current research indicates that the pathophysiology of OCD is associated with GMV abnormalities not only in corticostriato-thalamo-cortical (CSTC) circuit, but also in the cerebellum.

ABSTRACT

OBJECTIVETo investigate the associations between the drug responses to obsessive -pulsive disorder (OCD) and six functional genes related with serotonin and dopamine.

METHODSOne hundred and thirteen OCD nuclear families were collected. The OCD patients were treated with serotonin reuptake inhibitors (SRIs) for 8 weeks and the drug responses were assessed using the Yale-Brown obsessive-compulsive scale (Y-BOCS). The patients were divided into drug responders group and non-responders group according to the reducing rate of Y-BOCS score. The genotypes of six genes were determined with the Amp-FLP and Amp-RFLP techniques and analyzed by transmission disequilibrium test (TDT). The six genes are serotonin 2A receptor (5-HT2A), serotonin transporter (5-HTT), dopamine D2 receptor ( DRD2), dopamine D4 receptor (DRD4), catechol-O- methyltransferase (COMT) and monoamine oxidase A (MAOA).

RESULTSNo association was found between the six genes and different drug responses groups. However, there was significant difference between the drug responders and non-responders in homozygosity at the 5-HT2A -1438G/A locus (chi(2)=4.69, P=0.03).

CONCLUSIONThe results suggested that the 5-HT2A may play some roles in the effects of drug treatment on OCD.

ABSTRACT

Objective To investigate the shape and F-actin of human periodontal ligament fibroblasts(HPDLF) under different tension stress in vitro so as to learn the stress-biological effects of HPDLF. Methods HPDLF were cultivated for 6 passages and observed morphologically and identified by immunocytochemistry to be positive anti-vimentin and negative anti-keratin. HPDLF were divided into four groups: control (without tension stress), static tension stress (5 kPa), dynamic tension stress group 1 (5 to 0 to 5 kPa at the frequency of 3/min) and dynamic tension stress group 2 (5 to 2.5 to 5 kPa at the frequency of 3/min). The cells of each group were observed at the different time points of 16, 24, 32 h. The projection areas and shapes of cells as well as the structure of F-actin were examined by laser scanning confocal microscope and immunity fluorescence technique. The relationship among tension stress, time, shape and the structure of F-actin of HPDLF was analysed. Results In the dynamic tension stress group 1 and 2, the shape and the arrangement of F-actin of some HPDLF underwent regular changes at 16, 24 h, but the changes appeared to be obvious at 32 h. In the static tension stress group, it was found the increase of interspace of HPDLF, but no obvious changes in the structure of F-actin. Conclusion Different tension stress patterns had different effects on the shape and F-actin of HPDLF. Especially in the dynamic tension stress group 2, it showed direct ratio between the cell projection areas, the average fluorescence density and different loading time.

ABSTRACT

Objective To test the fracture loading of extracted cracked teeth with CEREC CAD/CAM ceramic inlays/onlays. Methods All the extracted teeth were divided into two groups (A: control group; B: test group; n=15 teeth in each group). All the teeth were prepared into those imitating cracked teeth. Those teeth in group B were strengthened with CEREC CAD/CAM ceramic inlays/onlays. All teeth were thermocycled and mounted for testing and then were loaded until fracture. Results The force of anti-fracture of extracted teeth imitating cracked teeth with CEREC CAD/CAM ceramic inlays/onlays (group B) was obviously higher than that of the teeth without CEREC CAD/CAM ceramic inlays/onlays (group A). Conclusion CEREC CAD/CAM ceramic inlays/onlays can effectively prevent the crack of teeth in clinical practice.
